A therapeutic protein-subunit vaccine from Genocea Biosciences for people already infected with HSV-2. In Phase 2 trials it reduced viral shedding and genital-lesion rates for up to 12 months. Despite these results, Genocea halted the program in 2017 to pivot to cancer immunotherapy, and it has not been revived by any developer since.

What it is

GEN-003 was a therapeutic vaccine — one meant to help people who already have HSV-2, not to prevent infection. It was a protein-subunit vaccine, meaning it used purified pieces of the virus rather than the whole virus or its genetic code. Specifically it combined a fragment of HSV-2 glycoprotein D (gD2ΔTMR) and a fragment of the viral protein ICP4, together with Matrix-M2, a saponin-based adjuvant (an ingredient added to strengthen the immune response).

Where it stands

GEN-003 reached Phase 2 (NCT02114060). In its dose-optimization trial, the better-performing doses reduced HSV-2 viral shedding and genital-lesion rates, and some of that effect persisted at 12 months. On its own terms, this was one of the more encouraging therapeutic-vaccine datasets in the field.

Despite that, Genocea Biosciences discontinued GEN-003 in September 2017, redirecting the company toward cancer immunotherapy rather than funding the large, expensive Phase 3 that would have been needed. No other developer has taken the program forward, so as of the date above it remains halted. GEN-003 is a useful reminder that a promising mid-stage result is not the same as an approved product — programs can end for business reasons, not only scientific ones.
